Cytotoxicity of PLGA-zinc oxide nanocomposite on human gingival fibroblasts

Background. Polylactic-co-glycolic acid and zinc oxide (PLGA-ZnO) nanocomposite has been investigated for its antibacterial properties, which could be beneficial adding to wound dressings after periodontal surgery. However, cytotoxicity against human gingival fibroblasts (HGFs) remains unclear should evaluated. Methods. ZnO nanoparticles were synthesized using the hydrothermal method. These metallic incorporated into PLGA matrix by solvent/non-solvent process. The nanomaterial was evaluated field emission scanning electron microscopy (FESEM), Fourier transform infrared (FTIR), thermogravimetric analysis (TGA), x-ray diffraction (XRD) analyses. HGF cells acquired from National Cell Bank categorized four groups: ZnO, PLGA, ZnO-PLGA, control. exposed different (1, 20, 40, 60, 80, 100 µg/mL) (0.2, 4, 8, 12, 16, 20 concentrations 24 48 hours. tested MTT assay. data analyzed SPSS 25, P<0.05 considered statistically significant. Results. exhibited significant toxicity at≥40 µg/mL viability decreased significantly at all hours of exposure. PLGA-ZnO cell in similar control group up 80 µg/mL. Conclusion. toxic high with prolonged Therefore, incorporating a biocompatible polymer such as strategy reducing their toxicity.
